3-Aryl-oxetanols are valuable building blocks featuring an aryl group and a hydroxyl group attached to a four-membered oxetane ring. Their distinctive structural features make them attractive scaffolds in medicinal chemistry and drug discovery, particularly for exploring bioisosteric replacements of carbonyl-containing functional groups.
The hydroxyl group can undergo reactions at different sites to give ester or ketone bioisosteres. For example, Brønsted acid catalysis selectively activates the tertiary benzylic alcohol of 3-aryl-oxetanols, which reacts with simple alcohols to form ethers that can serve as ester bioisosteres.

Alternatively, substitution of the hydroxyl group can give 3,3-diaryl oxetanes as ketone bioisosteres. In this process, elimination of the hydroxyl group can generate a carbocation, which subsequently undergoes a Friedel-Crafts reaction with an arene, replacing the hydroxyl group with an aryl group.

Beyond functionalization at the hydroxyl-bearing carbon, the oxetane ring itself offers opportunities for further transformation. For example, chiral phosphoric acid-catalyzed desymmetrization of suitable 3,3-disubstituted oxetanes enables enantioselective ring opening to access chiral vicinal diols. (https://lnkd.in/ebVygMEt)
