Aryl bromide-containing amino acids combine the intrinsic reactivity of amino and carboxylic acid groups with an additional aryl bromide handle, providing a broader range of synthetic possibilities than conventional amino acids.

The carboxylic acid group can undergo condensation with amino groups to form amide bonds, including peptide bonds when amino acids are involved, or with alcohols to form esters.

The amino group can participate in condensation with carboxylic acids to form peptide bonds or undergo C–N bond-forming coupling with halides and other suitable electrophiles. The aryl bromide moiety provides an additional versatile reaction site, enabling C–C bond formation through coupling with boronic acids, halides, and related partners, as well as C–N bond formation through coupling with amines.

The combination of multiple, orthogonal reaction sites makes aryl bromide-containing amino acids useful building blocks in medicinal chemistry and peptide-inspired research. Their diverse reactivity enables the construction of structurally diverse cyclic peptides and peptidomimetics. For example, aryl bromide-containing amino acids are utilized in the synthesis of #Daraxonrasib and #RMC7977, demonstrating their utility in the synthesis of structurally complex molecules.
