1,2,3-Oxathiazolidine 2,2-Dioxide Building Blocks for Boc-Aminoethylation

1,2,3-Oxathiazolidine 2,2-dioxide is a five-membered O-S-N cyclic sulfamidate bearing an electron-withdrawing SO₂ group.
1,2,3-Oxathiazolidine 2,2-dioxide is a five-membered O-S-N cyclic sulfamidate bearing an electron-withdrawing SO₂ group.
Its Boc-protected derivative is a high-performance
Boc-aminoethylation reagent widely used in medicinal chemistry.
The strained ring serves as an internally activated electrophile: nucleophilic attack induces ring-opening with SO₂ extrusion, delivering Boc-aminoethyl groups efficiently under mild conditions.
This reagent eliminates over-alkylation drawbacks and harsh reaction conditions seen with conventional ethylating agents. It also supports efficient N-alkylation of challenging heterocycles, facile construction of piperazine/morpholine fused rings, and stereoselective incorporation of chiral aminoethyl moieties.
As an indispensable alkylating building block for modern drug discovery, it is fully developed at Chemscene. A broad selection of derivatives is available, with select products scalable to hundred-kilogram quantities.
